1- Inorder to create a tomato variety that is resistant to frost damage we need to carry out the following steps.
i- We need to isolate the gene from frost resistance from the plant which is grown in the first composition.
ii- We need to clone this gene into the binary vector which has antibiotic resistance gene. which helps in screening the colonies which have our gene of interest. Binary vector also has the Left T-DNA border and Right T-DNA border which will help the gene to stably integrated into the plant genome.
iii- Then transform the agrobacterium tumefaciens which have the helper plasmid which will help the frost resistance gene to stably integrate into the tomato genome. Helper plasmid encodes the vir protein which encodes the protein which helps in the integration of DNA into the plant genome
iv- Transform the bacterium on the isolated plant cell. Plant cells which have infected with the bacterium will be able to grow on the antibiotic plate whereas normal cell will not be bale to do so.
a- for the cloning we will use T-binary plasmid and vir plasmid.
b- for isolating the salt tolerant gene we need to grow the plant which has the capability of salt tolerance and isolate the mRNA of the plant cells. mRNA of the salt-tolerant gene will be abundant which helps us in isolating the gene. By comparing the expression profile of plant growing in salt tolerant condition and normal condition we can make the gene which is required for the salt tolerant condition.
Cloning steps will be same as that we use for Tomato frost tolerant gene.
2- From the given gel picture and outline of the gene we can make out that in the mutant, the restriction enzyme site is lost between 1 and 2 fragments as result of that we only got one fragment of size 58 bp on the gel
