,Q6 (4pts) Describe the use of the culture collection in interpreting an uncultured 16S rRNA sequence /Q7 (5 pts) If you were examining the changes in cyanobacterial activity that occurs between day and night would you use A) cloning sequencing and phylogenetic analysis of 16S rRNA genes: B) metagenomics C) metatranscriptomics; clearly explain why and what you would expect to find in the data Q8 (5pts) Pretend that you are an environmental microbiologist employed by a consulting firm hired to create a bioremediation plan for an aromatic carbon pollutant in soil at a former industrial site. As a first step your manager has asked you to isolate bacteria from the site's soil that would be capable of degrading the pollutant Describe the microbial growth media you would design and the steps you would take to enrich and isolate a pure culture capable of degrading the pollutant. Describe why you would take each stepExplanation / Answer
6. The uncultured collection of 16S rRNA sequence need to be obtained after applying modern methods of molecular biology like sequencing/next gen sequencing. If we obtain the sequence to understand evolutionary relationship amongst various bacteria, the results are difficult to comprehend. But if one is able to enrich the organism through several specific media growth methods, one might be in a position to obtain relatively pure culture for the sequencing. If one obtains sequence of pure culture then based on the sequences, one can easily detect the sequences that we need to understand based on homology or sequence similarity.
7. One should use meta transcriptions as it gives a comprehensive idea about the expression levels of various genes( mRNA activity) as well as the relative abundance of the genes being expressed during day time as well as night time. The results will be vast that need to be analysed carefully.
8. Initially obtain the soil sample. Then streak it on an agar plate containing general media like peptone and yeast. Either one should know which bacteria to enrich. Based on it one can grow that bacteria in a specific media. Species like Alcaligenes, Acinetobacter, Rhodococcus, Nocardia generally can degraded aromatic compounds One can systematically try enriching them one by one on different types of growth media(based on literature). Make sure to get a glycerol stock of the bacteria present on the site
If one is not aware of the bacteria,
