Cloning and 16S rRNA genes are used for bacterial identification, classification, and phylogenetics related research.

Metagenomics is not useful because total genome remains unchanged during favorable as well as unfavorable condition. So, we cannot predict genes related to that phenomena using genomics.

Metatranscriptomics will work fine because transcriptome data is the mRNA data which are transcribed from DNA and the proteins are expressed in that specific condition. If we compare the transcriptome of day and night of the cyanobacterial colony we can acquire details about genes involved in that incident and their mechanism and probable pathway of action.
