Question
PLEASE HELP! 10. Estimate the size of the PCR product in the gel image below. Using the details about the EcoRI cut sites in the pCR4-TOPO vector that you used for the TA' cloning experiment (see Laulima, Lab 9... >Lab Protocols> PCR4TopoTA.bmp"), approximate the expected base pair (bp) size of the fragment that EcoRI will generate in the digest of your recombinant plasmids. (3 points) MILU maker Figure 1 . Agarose gel electrophoresis ( 1% agarose) of PCR amplified product from barcoding sample "C" using COl-Marine PCR primer sets. Lanes 3-6 are four volumes of the same PCR product (10ul, 7.5ul, Sul, and 2.5ul). Lanes 2 &7 are positive and negative controls, respectively. Lanes 1 & 8 are Bionexus Hi-Lo 10kb Marker/Mass Ladder. Gel was stained with GreenGlo, and ran at 90V for 70 min. Gel image taken from MANT group, sec. 2Explanation / Answer
As the Bioneux 10kb marker resolve on 1% agarose gel shows it is the 400 bp size of PCR product. because the Bineux resolve as below 50, 100, 200, 300, 400, 500, 750, 1000, 1400, 1550, 2000, 3000, 4000, 6000, 8000, 10000 bp.
Therefore as per the Bioneux Hi Lo 10 kb marker the given PCR product size is of 400 bp
