P53 gene is involved in the regulation of transcription and it is a tumor suppressor gene and mutation in both of the copy of tumor suppressor gene leads to loss of function mutation.It leads to development of bone cancer,breast cancer and brain tumor.

2)Gain of function is related to protooncogenes.Protooncogene are the normal gene which due to gain turns into oncogenes which leads to development of cancer.Mutation in one of the copy of Protooncogene leads to gain of function.Loss of function mutation is present in tumor suppressor genes.These genes inhibit the formation of tumor.When there is loss of function mutation in both the copies of gene then it leads to occurrence of tumor or cancer.
