1. Describe the selective process of protein turnover which involves ubiquitim asing an example of where it regulates a cell proce marks). I have covered some examples in lectures but you should find one different example and cite your source (s) (25% marks) lease answer this question using no more than one A4 page per question (potsjncluding any diagrams or Tables) you may wish to use. Hi Answer:
Q. Can you please give an example about the ubiquitin and proteasome
Answer: The Ubiquitin is the very small eukaryotic protein having a molecular weight of 8.5 kDa. It was first discovered and characterized by Gideon Goldstein in 1975. Studies revealed that the mammalian and yeast ubiquitin have 76 residues and they differs from each other by two residues only. The major genes which code this protein in human genome are UBB,UBC, UBA52 and RPS27A. The ubiquitin protein carry out the process known as Ubiquitination which affect the various cellular processes by protein degradation regulation in the cells. The most common example of Ubiquitin is: E3 ubiquitin-protein ligase SMURF1, which regulates glucose metabolism and also shows the inhibitory action on osteoblast differentiation process. ATP-dependent proteolysis factor 1 (APF-1) is also considered as the ubiquitin as it has similar function to the ubiquitin.
Proteasome: are the protein complexes are the second intracellular degradation mechanism found in all eukaryotes and archaea and also in certain bacteria. The proteasome is the part of degradation mechanism of the cells and it degrade the unwanted and misfolded proteins and maintain the concentration of particular proteins in the cells. The protein which are undergo degradation by the proteasome are tagged by a unique small protein known as ubiquitin and this whole complex is known as ubiquitin-proteasome system (UPS). The UPS consit numver of enzymes and proteins. Various ligases and deuboquitins are currently used to break this UPS so that the degrdations of the proteins inside the cells can be checked. Example is 26S Proteasome.
