You are provided with the sample containing a mixture of 3 nucleic acid polymers. Polyacrylamide gel electrophoresis of the sample showed three bands: one band proceeded quickly to the head of the gel, a second band traveled midway between the origin and the front, and the third band moved only slightly from the origin. SDS gel electrophoresis of another aliquot of the same sample showed only one band. Comment on the degree of super coiling and whether the DNA in each nucleic acid was more likely linear or circular.
Explanation / Answer
In electrophoresis the DNA moves on the gel according to size, smaller the size faster it migrates and same sized DNA with linear DNA migrates slower than circular DNA and it moves slower than super coiled form of the same sized DNA due to the porosity of gel .
In SDS gel the DNA moves according to size only so it has all three different forms of the DNA with same size at same band .
