DNA PURIFICATION QUESTIONS:
Why is it important to remove as much as possible of the growth medium from the cell pellet during cell harvesting?
Alice and Frankie are lab partners in a micro lab performing DNA extraction, each person using a different bacterial species. Both of them spun 1.8 ml of their overnight broth cultures in a microcentrifuge, and it turned out that Alice’s tube had a substantially larger pellet than Frankie’s. What could Frankie do to increase the amount of cell pellet necessary for subsequent extraction steps?
Explanation / Answer
Growth medium contains antibiotics and lot of cell debris and proteins and waste produced from the cells during growth of cells. So after cell harvest and used in DNA extraction or RNA isolation this debris make problem with isolation.
Increasing the extraction volume from the culture then centrifuge to increase the amount of cell pellet. Because volume of culture is directly propotional to amount of cells.
And also due to the bacterial species grown by Frankie has high doubling time the growth rate will be low it takes long time to reach the same amount of cells compare to Alice bacterial species.
