Question
Lysozyme was assayed at bacterial cell wall, [S] = 2.5x10-8 g. The Km for the substrate = 6x10-3g. At the end
of one minute 2.5% of the cell wall hydrolysed. Answer the following questions using this information: (9
points)
a) What percent of bacetrial cell wall will be hydrolyzed at the end of 3 minutes assuming product formation
is in 1st order of reaction?
b) What will be a concentration of hydrolyzed product at the end of 4 minutes?
c) Determine maximal velocity (Vmax) with the enzyme concentration used.
Explanation / Answer
a) Given, at the end of 1 minute, 2.5% of the cell wall is hydrolyzed. So, at the end of 3 minutes, cell wall hydrolyzed = 2.5 x 3 = 7.5%
b) At the end of 4 minutes, the % of cell hydrolyzed = 2.5 x 4= 10%.
Initial enzyme conc = 2.5E-8g
So, at the end of 4 minutes, conc is = 10% of 2.5E-8g = 2.5E-9g
For part c, I need the initial velocity.
