(a)When measuring with a pH electrode, several things limit the accuracy of the measurement. In each of the following situations, describe what would cause the observed effect:

(b) You are trying to measure the pH of a biochemical system at –in situ temperatures (37.5 °C). Before your experiment, you calibrate the instrument using stock room temperature buffers you pulled from the cabinet. However, your measurements are consistently 0.25 pH units greater than predicted by the literature

Explanation / Answer

Not vary sure for biological system, however just to help you some explaination. Please cross check which direaction you are looking for

1) Many of the times CO2 in air is absorbed by solutions which forms carbonic acid because of which we get higher pH. Thats the reason any freshly prepared DI, distilled, RO water over a period of time shows acidic pH

Other reason may be

2) effect of pH measurement temperature on electrode (effect of temp on electrode membrane permeability)

3) Also may be think of the effect of measurement temerature on the biological system
