3. The first and the prior method to distinguish the bacteria is gram staining technique, by this method we can identify the bacteria either gram positive or gram negative. Gram positive bacteria have more peptidoglycan in cell wall than gram negative bacteria. Acid-fast staining is used to identify the mycobacterium sps., and endospore staining, flagellar staining, negative staining are used to detect the spore formers, motility of the bacteria and presence of capsule or not respectively.
