Question
absorbance vs log dna bp plot, with interlacation
I already did the experiment and have the original absorbance vs log dna bp graph done with ethidium bromide and 19 different concentrations of DNA. I was able to determine the amount of bound ethidium , but am unsure of what to add to my excel sheet with this question. any help would be great
The anticancer drug actinomycin D (act D) has a high affinity for intercalation at the sequence GC. Sketch the absorbance versus log IDNA (bp)] plot for the addition of ethidium bromide to a solution of herring testes DNA for which all GC sites have been bound by act D. Indicate how the plot would differ from the one obtained in your experiment without having act D present in the system.Explanation / Answer
Without having act D in the system, the GC sites would be more exposed to the ethidium bromide. This shows that the slope of A0 vs log bp would be more and hence the nature of the plot increasing. Rate me for further clarifications
