Question

You want to fill a culture chamber (surface area = 10 cm2) with freshly dissected cells at a plating density of 50,000 cells/cm2. In the culture chamber, you also need to put in 10 mL of total volume so cells have enough nutrients to begin with. Now, let's assume that you have prepared a concentrated cell suspension at 1 million cells per mL. How much volume of this concentrated cell suspension do you need and how much volume of plain culture medium (i.e. without cells) do you need to give you the necessary number of cells required for the culture chamber per the recommended plating density and 10 mL total volume? Your answer should be two volumes, one for cell suspension and another for plain culture medium.

Please I need Help with this problem

Explanation / Answer

since you need 50000cells/ cm2 in an area of 10cm2 using liquid of concenteration 1million cells/ml

Volume of concenterated cell suspension = 50,000*10/1,000,000=.5ml

Since total volume =10ml the volume of culture medium = 9.5 ml
