You want to determine if a bacterial species has a similar DNA sequence to another bacteria. The sequence may also serve as a replicator.You isolate genomic DNA from the species and cut the DNA with restriction enzymes separate the DNA on a gel and transfer it to a membrane for Southern Blot analysis using a newly identified replicator sequence as a radiolabled probe
The first trail leaves a blank membrane with no signal even in the sample from which the DNA was originally identified (positive control showed no signal)

The samples was simply to highly concentrated with DNA samples of different lengths. This issue rises from not uses a centrifuge or some machine to separate the different lengths of the DNA strands. Since so many different length strands were in that one sample, they all diffused at different rates and just left black marks all over the first trial. Also due to the high concentration the black mark was thick and continuous
