Question

Prostaglandins are a class of eicosanoids, fatty acid derivatives variety of extremely potent actions on vertebrate tissues. Prostaglandins are responsible or producing fever and inflammation and its associated pain. They are derived from the 20-carbon fatty acid arachidonic acid in a reaction catalyzed by the enzyme prostaglandin endoperoxide synthase. This enzyme uses oxygen to convert arachindonic acid to PGG2, the immediate precursor of many different prostaglandins. (If you use the computer for this analysis, email me your file to assist with awarding partial credit, if desired.) The kinetic data given above are for the reaction catalyzed by prostaglandin endoperoxide synthase. Determine the V_max and K_M of this enzyme. Ibuprofen is an inhibitor of prostaglandin endoperoxide synthase. By inhibiting the synthesis of prostaglandins, ibuprofen reduces inflammation and pain. Determine the type of inhibition and the inhibition constant (K_i and/or K_i, as appropriate).

Explanation / Answer

Vo = Vmax [S] / Km+[S]

44 = Vmax [3.5] / Km + 3.5..................1

32.2 =Vmax [1] / Km + 1........................2

Solving eq1 and 2, Km = 1.77 mM

Solving for Vmax: - Put the value of Km in equation 2.

32.2 = Vmax/ 2.77; Vmax = 2.77 * 32.2 = 89.3 mM/min.
