Question
Describe a good way to graphically define Tm and determine Tm for this graph. Graphically define and determine the half-width (half-width at half-maximum), ?T, of the melting transition from the derivative plot. Describe how you deal with the “offset” of the derivative curve—the fact that dD/dT never reaches zero. Briefly discuss the physical meaning of this width. For example, what sorts of membranes will give narrow and wide widths. (Remember that these data are “made-up” data that only roughly correspond to real membranes.)
Explanation / Answer
Tm for this graph is around 55 degree C. Increase in temperature increases membrane fluidity.
Membranes which are more fluid will give wide width, while the membranes which have greater number of unsaturated fatty acids will give, narrow width.
