The researchers were interested in sequencing the genome to determine if this new life form was similar to other single-celled eukaryotes. The researchers roughly purified the genomic DNA and decided to size fractionate the DNA using density gradient centrifugation.They got the following result when assessing the amount of DNA in each fraction from cesium chloride gradient. GC content of DNA determines the buoyant density of double stranded DNA. The buoyant density is determined by Cs Cl density gradient centrifugation. When the eukaryotic DNA is subjected to Cs Cl gradient centrifugation, 2 bands are formed.
1)Main Band-This corresponds to the genomic DNA of the organism. The genomic DNA appears as a broad band centred at a buoyant density corresponding to the average GC content of the genomic DNA.
2) Satellite DNA- corresponds to short tandem repeats that occur as large clusters around the centromeres in eukaryotic chromosomes. This DNA may or may not be transcribed but is never translated. The satellite DNA appears as small peaks that has lower/ higher buoyant densities in comparison to the main band (genomic DNA).
