Question

In order to begin a research project scientists often need tocreate or synthesize materials that can be used in research. In thecase of true research these materials often cannot be purchased. This is the status of the research project now being under taken inyour biochemistry course. You have been working with plasmidscreated previously by the research team working with Dr. March. Inorder to further this research it is essential to make materials(in this case protein expression plasmids) that will serve as abasis for further research.

We have accomplished this, BUT we need to be sure about ourplasmids.

Explanation / Answer

I can't be completely sure with one answer to this question with out knowing the plasmind. One easy way to determine if your cells have taken up the plasmids is if the plasmid-bearing cells have a competitive advantage over the plasmid-free cells. You can utilize selective pressure to select for the plasmid-containg cells. Its the most commonly used experiment when a plasmid is introduced that contains antibiotic resistance. You simple inoculate a agar medium containing the particular anti-biotic with you what you hope is your anti-biotic resistant strain and incubate it for a period of time depending on what microbe your using and see if you get any proliferation. Other forms of selective pressure forms can be a) deletion of a essential gene from the bacterial chromosome and its inclusion in the plasmid b) introducing a growth repressor in the baterial genome and its antidote in the plasmid. Hope this helps. If you thought that I was helpful if you don't mind please comment it helps me see how I am doing.
