The protein is made up of the thee subunits and they are joined by the sulpher bonds, when they are subjected with the 2-mercaptoethaol, there bonds get broken due to action of mercaptoethanol on the supher bonds. so these are separated on the SDS-PAGE into 3 different subunits.
