Question
DETERMINATION OF DNA RESTRICTION FRAGM 4 is a photograph of a typical student gel produced by the procedure outlined above. In this pho. tograph, lanes 1 and 6 have marker DNAs down to a couple of hundred base pairs at the bottom of the gel. Lane 2 DNA; lane 3, double digested DNA; lane 4, Eco RI digested DNA; and lane 5, an unknown restricti the lambda DNA in lane 5? If so, place your answer below and explain why you came gel has Hind III digested Lambda Lambda DNA digested with on endonuclease. Can you identify the restriction endonuclease which was used to cut with sizes ranging from 10,000 base pairs at the top of the er below andExplanation / Answer
Lane 5 contains DNA sample digested with HindIII restriction enzyme. We can say this because the lane 5 corresponds completely with the pattern of lane 2. Lane 2 has prominent bands at 2300 , 2000 and another near 500 base pairs. So does lane 5 which has unknown enzyme digested DNA sample. Moreover none of the bands correspond to EcoRI bands. Hence the unknown enzyme is Hind III
