Question
29. (6 points): You are a lead food scientist in a yogurt company and responsible for developing a new Streptococcus thermophilus strain that is resistant to two types of phages, A and B. Currently, you have S. thermophilus A strain that is resistant to phage A only AND S thermophilus B strain that is resistant to phage B only. Using your knowledge of CRISPR system, design your method to create a S. thermophiles strain that is resistant to both phages? You are free to either modify the existing A or B strains or introduce/engineer a new S. thermophilus strain. No need to write technical details such as cloning or transformation. I just want general description in 4-7 sentences. You can use a diagram as part of your answerExplanation / Answer
To make a new S. thermophilus strain that is resistant to both A & B types of phages we need to incorporate both genes that make them resistant in a single strain. For that purpose we need to first isolate the resistant gene from one organism (It can be strain A or strain B or a new strain that is resistant to either phage A or phage B. For theory we say that we isolate gene A). Once the gene A is isolated we have to insert this gene in S. thermophilus strain B. We can do it by CRISPR technology. We will transfected the cell with an enzyme complex containing an artificial guide, Cas9 enzyme & the isolated resistant gene A. Artificial guide will help to find the targeted site in DNA. Then Cas9 enzyme will cut the targeted site & then new gene (gene A) will inserted in the targeted site & ultimately cell will repair this cut. In this way we can generate a new S. thermophilus strain that is resistant to both A & B types of phages.
