Adapted from The American Society of Human Genetics i eut Measüring Gene Expression Cardiac myocytes and neurons from the same person have different phenotypes because of differences in their gene expression profiles Tanning is the result of skin cells responding to the environment by changing their gene expression profile. How do you think scientists originally tested the ideas that differences in gene expression explain phenotype differences among cell types and among cells of the same type in response to the environment? Come up with an experimental design, including appropriate controls, not technical details or specific techniques Address the following in you plans 1. experimental plan to test one of the hypotheses. Focus on general . What samples will you collect? Will you repeat the experiment? How? What molecule(s) will you look at to see if there are differences in gene expression? What about that/those molecule(s) will you measure? Are any controls necessary? . 2. Complete the DNA Microarray Virtual Lab: http/leam.genetics utah.edu/content/labs/microarray 3. Answer the following questions on a separate sheet of paper In order to measure gene expression with a microarray, what molecules do you need to isolate from your sample? How are the isolated molecules modified before the sample is added to the array? Why? What do the spots on a microarray contain? Are the molecules with a single spot the same or different? What about the molecules in two neighboring spots? What property of DNA does microarray technology take advantage of? Scientists must be very careful to scan their microarrays in a certain orientation. Why is it bad if they scan the array in the wrong orientation? - Using a microarray, woul ld you be able to measure gene expression from a gene if the sequence of that gene is not known? y, would you be able to detect differences in gene expression between two Using a microarra samples that affect the level of functional protein made, but not the level of mRNA? - In the example in the virtual lab, why is it important to compare skin cancer to healthy skin cells specifically? For example, why can't you compare the gene expression of skin cancer cells too healthy blood cells? -

Explanation / Answer

1)

The central dogma states- DNA goes to RNA goes to polypeptide. Hence which ever genes are going to be expressed will have RNA transcribed. So we will sample RNA. We can extract nucleic acid and use DNA digesting enzymes to destroy DNA. We can compare the cells of different location from same individual for RNA profiles.if we are looking for a particular gene, we can use northern blotting to transfer the RNA to membrane and check for the gene using radio or fluroscently labeled probes. The controls can be the RNA samples from different cells or even DNA as DNA sampling will bring all the genes into the profile and a comparison can tell us about the missing ones.
