Question
a) What happens to a previously assembled actin filament that is placed in a test tube containing 0.5?M ATP-G-actin solution and is incubated there for one minute (you can assume that the solution contains an excess of ATP and Mg2+)?
b) The experiment in (a) is repeated, but this time with a starting concentration of 50nM actin filaments and 0.5?M ATP-G-actin (and excess of ATP and Mg2+). What happens to the actin filament?
Many thanks.
These were all the information provided to me, followed by the following two questions:
c) The experiment in (a) is repeated again but this time a Cofilin molecule is included in the reaction. What happens to the actin filament?
d) How can blood platelets contain a 200?M concentration of unpolymerised G-actin?
Explanation / Answer
A) excess of ATP in the excess amount of bivalent cation Mg2+ ,actin filaments shows enhance stability with conformational changes. Because filaments polymerization is needed bivalent cation which tightly bind to ATP and helps in polymerization in free monomer of ATP - G -actin.
B)here actin filament is 10 times lower than metal free actin monomer which in excess of Mg2+ and ATP polymerize actively and very fast and hydrolysed from other end to make balance between monomer and polymer.
