the differential stains are those stains which differentiate between two organisms by using two different staining dye.
in case of gram staining, it differentiates the organisms into gram +ve and gram -ve.
In this procedure the organism is fixed on the slide by heating and after that crystal violet dye has been applied. after washing with water grams Iodine is applied and this makes a complex with stain and forms crystal violet-iodine complex. after that slide is washed with water and alcohol. after washing counterstain safranin is applied and after some time slide is washed with water and viewed under the microscope.
the basic principle behind this is gram +ve organism contains peptidoglycan layer and less amount of lipid in which the first stain enters and forms a complex with Iodine solution and trapped inside. now when we wash with alcohol then organism contains large amount of lipid loses this complex and became colorless and when counterstain applied they again take the color and became red.
