Question

I already know the answer is C. I need to know the exact steps in how to solve this questions

17 to 21 based on the following information. The enzyme urease catalyzes the decomposition of urea into ammonia and CO2. The following values of urease have been determined at 10 nM total enzyme concentration, i.e., [E]T = 10 nM. KM = 25 µM; kcat = 1x104/s Hint: 1 µM = 10-6 M; 1 nM = 10-9 M

18) The intercept at the (1/S) axis of the Lineweaver-Burk plot of urease is ___.

A) 0.04/µM B) 25 µM C) -0.04/µM D) – 25 µM

Explanation / Answer

since V= VmaxS/(KM+S)

1/V= (KM/Vmax)*1/S+ 1/Vmax

this is in the form of y= mx+C, x intercept is obtained by putting y=0, mx+C=0 and x = -C/m

for the case given intercept is -1/Vmax/(KM/Vmax)= -1/KM

The intercept of 1/S is -1/KM

=-1/25 = -0.04/uM ( C is correct)
