A promoter sequence of a bacterial DNA shows -10 and -35 regions. Promoter sequences could be seen on DNA. The regions -10 and -35 are consensus sequences in a promoter. These consensus sequences shows sequencely aligned frequent residues. -35 is TTGACA. -10 is TATAAT. Both -10 and -35 boxes could be seen at 10 bases and 35 bases upstream of transcription start site (+1). At these consensus sequences, the transcription factor binds to initiate mRNA transcription . Transcription factors are protein activators/repressors. The -10 and -35 concensus sequences can be identified only by RNA polymerase.
