Question

You found a gene that is predicted to encode a 30kDa--Protein J. After generating a highly specific antibody, you perform a western blotting analysis on Protein J. You found a single band that displayed ~27 kDa rather than at 30 kDa. The sequence analysis indicates that the very N-terminal of Protein J is composed of a stretch of hydrophobic/non-polar amino acids. Which if the following is most likely CORRECT for Protein J?

A. Protein J is a soluable ER lumen protein

B. Protein J is produced inside of mitochondria

C. Protein J intercts with Protein H

D. Protein J is produced on smooth ER surface

Explanation / Answer

Answer - A. Protein J is a soluable ER lumen protein.

After protein enter into the ER lumen N-terminal of protein is cleaved by peptidase present in the ER lumen. N-terminal of protein rich in hydrophobic/ non polar amino acids it makes a transmembrane alpha helix in intermembrane space of ER. The remaining part enter into the ER lumen then peptidase cut the N-terminal. The function of the rpotein is not affect by this process.
