Question

Name SET 1 (sopts) 2018 vou are studying the glycosylation of a membrane SFSU ID osy pl. The N- and C-terminal domains have been ciroled protein that has the tollowing N-term C-term 50 100 150 200 250 300 350 400 amino acid number A. Draw the predicted structure of the protein in the plasma membrane, including transmembrane and soluble domains. CLDH Plasma membrane You conclude that either the N-or the C-terminal domai not both. Please explain the reasoning behind this conclusion. B. contains a glycosylation site, but mino ao -lycosidle will not Farm on menionine an C. What is one example of another piece of information you need about the protein to determine whether the N-or C-terminal domain carries a glycosylation site?

Explanation / Answer

C. As seen in the previous part, this is a transmembrane protein with amino terminal and carboxyl terminal on either side of the membrane. Another piece of information is whether the membrane protein is extracellular or intracellular or transmembrane.

As is seen this is not a secreted protein. Neither is it extracellular or intracellular as it has seven transmembrane domains. As N-terminal is not likely to have glycosylation site due to presence of methionine, it can be concluded that carboxyl terminal of the protein is most likely to have N-linked, O-linked or C-linked glycosylation site.
