Question

C. Moving between the gene in the genome and the mature mRNA product is a difficult endeavor for some students. Let's start with the following figure from your book. 5 3 3' Completed RNA transcript Answer the following questions. 1. Is the "completed RNA transcript" the pre-mRNA or the mature mRNA? What made you answer they way you did? 2. Where is the TATA box sequence on this figure (one or more of a-f)? Is it in the DNA only, the RNA only, or in both? 3. Where is the 5'-UTR sequence on this figure (one or more of a-f)? 4. Where is the sequence that encodes the 5-UTR on this figure? (one or more of a- 4. Where is the terminator sequence on this figure (one or more of a-f)? 5. Where could a nucleobase sequence corresponding to the 100th codon in the protein coding sequence be located in this figure (one or more of a-f0?

Explanation / Answer

1) It is mature RNA because completed RNA transcript is smaller than the original template, RNA splicing is the process that occurs in the cells. In RNA splicing, non coding parts i.e. introns of pre-mRNA, are recognized and removed by a spliceosome.

2) b is the TATA box sequence present in eukaryotes and archaea. In Prokaryotes TATA box is called the Pribnow box that consists of the six nucleotides TATAAT. it is considered as non-coding DNA sequence present in promoter region and facilitate the binding of RNA polymerase.

3)and 4) a and e is the 5' UTR. 5'UTR is the region present in mRNA upstream to the initiation codon. Sometimes the portion of the 5 UTR translated into a protein. The translation of mRNA main coding sequence is regulated by this product. 5 UTR is completely untranslated in many organism, instead of forming secondary structure complex to regulate translation. The 5 UTR interact with proteins concerned to metabolism; and proteins translate sequences within the 5 UTR. also, this region has been participated in transcription regulation, like sex-lethal gene in Drosophila.

5) d and f is the terminator sequence. it is the nucleotide sequence present in the DNA template serve as a signal to stop transcription. RNA pol detaches and RNA synthesis ends.
