Question

While the generation of stably transfected tissue culture cells is a major experimental approach, over time some cell lines lose the ability to express the transgene. This process of gradual silencing is known as extinction. Analysis of some of the cell lines with transgene extinction indicates that there is no change in the nucleotide sequence of the gene (including regulatory sequence and untranslated sequence) and no change in location (chromosome rearrangement has not occurred). State a hypothesis to explain the process of extinction.

Explanation / Answer

Explanation:- The most probable reason for extinction or gene silencing would be that the transgene might undergo transcription but the mRNA could not undergo translation due one or other factors thus polypeptide would not be formed thus causing extinction.
