Enlist the significance of following reagents: Calcium Chloride Antibiotics: Heat Shocks: Ethanol Treatment: SDS: Compare between efficiency of two pipettes: 20-100 and 20-200, if you expected to draw 50 microliter (mu l) of any solution. For three times you drew fluid, you observed that withdrew 52 mu l; 51.5 mu l; and 50.5 mu l. Subsequently, the withdrew 55 mu l; 53.5 mu l and 54.85 mu l. Calculate the average difference of observed minus expected weighs between the two pipettes. Which one is more suitable for use? Show your work. What will happen if you forgot to use Chloroform during DNA extraction?Explanation / Answer
5.a. Significance of Calcium chloride (CaCl2) :- In the transformation experiment in laboratory technique , It increases the ability of a prokaryotic cell to uptake of DNA in to cells, and incorporate plasmid DNA , which is used in the Recombinant DNA technology. It also stabilize and promoting the activity of certain enzymes, which is used in extraction procedure. The calcium chloride promotes the binding of plasmid DNA to lipopolysaccharides (LPS). Calcium ions are positively charged, they attract both the negatively charged DNA backbone and the negatively charged groups in the LPS inner core.
b. Significance of Antibiotic:- Antibiotics are used to kill specific target and cell. Antibiotics are used to prevent and in treatment of bacterial infections. They acts through the inhibition or regulation of cell wall synthesis, nucleic acid metabolism, and protein synthesis. Some antibiotics have been found to exhibit anticancer properties. They kill or inhibit the growth of bacteria.
c. Significance of heat shock:- They act like a chaperone. They are significant, in protein-protein interactions such as folding. They help in establishment of proper protein conformation (shape). They also prevent unwanted protein aggregation. They also activate Immune Response, because they delivers antigens from diseased cells to the immune system’s.
d. Significance of Ethanol treatment:-Ethanol used in alcoholic beverages, It is the most common antiseptic, used in antibacterial hand sanitizer gels and medical wipes. Ethanol denatures the proteins and kills the organism. It is effective against most bacterial and fungal infection.Ethanol also use as an antitussive agent. It also given as an antidote to methanol and ethylene glycol poisoning. It is a significant Fuel in some countries.
e.SDS:- Sodium dodecyl sulfate,(SDS) is a synthetic organic compound. It acts like a anionic surfactant. It is used in many cleaning and hygiene products, domestic cleaning, personal hygiene and cosmetic, pharmaceutical, and food products. It is also used in industrial and commercial cleaning and product formulations.
