The molecular weights of the bands in the gel are mentioned in the colored lane. It started from 10kbp to 250kbp.
The corresponding bands of the sample lane to that of the marker lane show that the sample DNA formed distinct bands at 10kbp, 15kbp, 25kbp, 37kbp, 100kbp and then at 250kbp.
All other bands are almost negligible. It indicates that 16ul of cell lysate consisted of
10+15+25+37+250+100 = 437kb
9.6ul of cell lysate consisted of (9.6*437)/16 = 262kb
