Question
For your research project, your group is planning to evaluate the diversity of static cultures inoculated with a bullseye strain of Pseudomonas fluorescens over time, in comparison to static cultures inoculated with a smooth strain. The experiment will be similar to the one documented in Figure 2 of the 1998 paper by Rainey& Travisano, with data collected in triplicate for each strain at each timepoint; however you're planning to plate samples from your cultures just on days 1, 2,3 and 4. How many static cultures of each strain will you need to start in order to collect the desired data after 1-4 days of growth? Enter just the number (for one strain) in the box below 1.0 0.8 5 0.6 0.4 0.2 0.0 1010 109 10' 0 2 46 8 10 Time (day) Figure 2 Effect of ecological opportunity on the evolution of genetic diversity. Two sets of identical populations were founded from the ancestral smooth morph and incubated either with or without shaking. Every 24 h, three replicate microcosms were destructively collected and diversity was determined by scoring the frequencies of morphs. a, b, Diversity increased rapidly in the heterogeneous environment (a) whereas no diversity was detected in environments lacking spatial structure (b).c, d, Evolutionary dynamics of the principal morph classes in the heterogeneous (c) and homogeneous (d) environments. Values are the means s.em. (n-3). Circles represent smooth morphs, triangles represent wrinkly-spreader morphs and squares represent fuzzy-spreader morphs.Explanation / Answer
Bacterial static liquid culture is not shaken, but the oxygen needed for the culture will be provided by the culture itself. Growth of the culture bacteria will follow the growth curve of four phases. The picture also shows the similar growth curve form. Diversity of bacteria is very vast as the structure and number of cells vary according to the density of the culture plated.
