Question

BIOL 3810 Molecular Cell Biology Lab Solution Making Problem Set 2 (10 pts) Name: 1. You are given a bottle of green liquid labeled "solution X". You want to make 5 ml of a 1/10 dilution of solution X. Describe the steps you would take to make this dilution. (1 pt) 2. You take the OD of the dilution and get a value of 0.35. Using the standard curve you constructed in class for the Fast Green solution, figure out the concentration of the dilution (from question 1) (1 pt) 3. Is this concentration (from question 3) lower or higher than the concentration of Solution X? Why do you say this? (2 pts) You try to take the OD of undiluted Solution X. The reading is off the scale (you cannot use your standard curve). How could you figure out the concentration of undiluted Solution X using just the information you have? (2 pts) 4.

Explanation / Answer

1) A solution of 1:10 dilution is made by mixing 1 part of the solution with 9 parts of water. Therefore, in order to make 5 ml of dilution 1/10 solution, 0.5 ml of the green liquid should be mixed with 4.5 ml of water.
