BIOCHEM
PLEASE ONLY ANSWER IF YOU TRULY UNDERSTAND IT!! I posted this many times, and people have been giving wrong answers. Also ONLY answer if you can answer it correctly and FULLY. Also a brief explanation helps! Thank you!
11) Name the following compounds. For each, identify if the base is in the anti or syn conformation and if it is a nucleoside or nucleotide.
12) What would be the sequence of the DNA strands that are complementary to the sequences below. Make sure the label the 3’ and 5’ ends. Which set of strand would be more stable upon formation of a double helix?
A) 5’-CGACTCGACCGTGC-3’
B) 5’-ACACTATAGAATGT-3’
13) Which sugar pucker do deoxynucleotides and ribonucleotides each adopt?
14) What are three uses of mononucleotides in biology?
15) What is the natural biological purpose of restriction endonucleases?
16) If you had a DNA double helix that had a Tm of 80°, what would its approximate melting curve look like? Does single stranded or double stranded DNA have a higher 260 nm absorbance? What could you do to further stabilize the helix (without changing the helix)?
17) Below is a sequence for a strand of DNA. If you sequenced it using Sanger Sequencing, draw bands in the box provided to show how your gel will look like? Hint- assume you have a 5’-radiolabeled primer that associates to the 3’ end of the strand.
NH2 O Nm HN HO O HO A) B) O OH OH O=P-0 O-P-O 2N NNIExplanation / Answer
11) Compound (A) is Adenosine. Name adenosine is due to adenine base. It is in anti-conformation.
It is a nucleoside (nitrogenous base covalently attached to a sugar but without the phosphate group).
Compound (B) is Uridine 3'-monophosphate. (Phosphate group on third carbon of the pentose ring). The Uracil base it in syn conformation.
It is a nucleotide (consists of a nitrogenous base, a sugar (ribose or deoxyribose) and one to three phosphate groups).
12) The two DNA strand in double helical structure of DNA are complimentary to each other, in which A always binds to T with two hydrogen bonds and C always binds to G by three hydrogen bonds.
(A - Adenine, T - Thymine, C - Cytosine and G - Guanine)
(A) 5’-CGACTCGACCGTGC-3’
Complimentary will be like: 3'-GCTGAGCTGGCACG-5'
(B) 5’-ACACTATAGAATGT-3’
Complimentary will be like: 3'-TGTGATATCTTACA-5'
Strand (A) will contain more number of CG base pairing. Bases C and G are binds through three hydrogen bonds provide more stabilization to DNA strand than A and T base pairing. Therefore strand (A) is more stable.
13) C2' endo conformation is adopted by deoxynucleotides in DNA and C3' endo conformation is adopted by ribonucleotides in RNA.
14) (a) Mononucleotides are building blocks of genetic material DNA and RNA.
(b) They also plays a central role in metabolism. They function to carry packets of energy within the cell in the form of the nucleoside triphosphates e.g., ATP, GTP, etc.
(c) They also participate in cell signaling.
