Question
Based on the figure above:
a) indicate which compounds can be visualized by using UV light during the TLC analysis. Indicate by stating Yes or No.
b) A 100 meters chromatography column was found to contain 20 theoretical plates. What would be the HETP of the column? (please show calculations)
c) What would be the HETP of a chromatography column that is 100 meters in length and contains 10 theoretical points? (please show calculations)
d) Based on part B and C, which column would give you the best separation? Briefly explain.
ex Iadicate which compounds below (9-13) can be visualized by using ultraviolet light (Le, UV) during TIC analysis. adicate your choice by stating Yes or No. 12 10 13 25-Compound 10 28-Compound 13 26 Compound il 24-Compound 9 27-Compound 12 chromatogapby columa (e-A) was found to contaia 20 theoretical plates. What would be the 29-A 100 meters HETP of the columa? (Show calculations)Answer Answer- columa (b) that is 100 meters in length and contains 10 30-What woald be the HETP of a chromatogapky theoretical plates? (Sbow calculations) Answer 31- Which columa would give you the best separation,a or s? Esxplain "hriefly why this is o Answer a or bExplanation / Answer
(b): Given the length of column, L = 100 m
Number of theoritical plates, N = 20
HEPT = L/N = 100 m / 20 = 5 m (answer)
(c): Given the length of column, L = 100 m
Number of theoritical plates, N = 10
HEPT = L/N = 100 m / 10 = 10 m (answer)
(d): More the number of theoritical plates(N) smaller is the value of HETP and higher is the separation. Hence the chromatography in part-(b) has better separation.
(a): alpha-beta unsaturated acid/ketone/ester/amine etc. compounds are easily detected by UV light during TLC. analysis. Hence compound that can be visualized are compound 11 and 12
