For these questions, assume you have been given the following information and answer the questions below: 5X TGB (Tris-Glycine Buffer) pH 8.3 Tris 25mM Glycine 250mM SDS 0.1%

2a) How many grams of SDS will you add when making 500mL of 5X TGB? (FW of SDS is 288.38)?

2b) If you were to make a 1X working solution of TGB, at what concentration will each component be? (2pts) Tris-base = _______________ Glycine = ________________ SDS = ____________________

2c) Using your 500mL solution of 5X TGB, show your calculations for making 100mL of 0.5X TGB.

Explanation / Answer

2a) 0.1 g x 500/ 100 = 0.5 g of SDS

2b) Tris-base = 5 mM Glycine = 50 mM SDS = 0.02%

2c) 100 mL x 0.5X = V mL x 5X

therefore V = 100 mL x 0.5X / 5X = 10 mL

i.e. 10 mL of 5X solution is tobe diluted to 100 mL for making 100mL of 0.5X TGB.
