A method for determining Vitamin D3 in blood plasma is as follows: 1.5 mL of plasma is combined with several reagents (2.0 mL standard solution, 0.5 mL methanol, 1 mL reagent solution, 400 uL KOH). This mixture is processed at 80.0 oC for 20 minutes, and then extracted into 10.0 mL heptane, centrifuged and dried. (The method assumes all Vit. D3 is extracted into the heptane.) The resulting dry powder is dissolved in 200. uL methanol/ water and analyzed. One sample analyzed in this way contained 1.2 ng/mL. Calculate the concentration of Vit. D3 as ng/mL in the patient’s plasma.

According to the method analysis of 1.5 ml of plasma we get 1.2 ng/ml of vitaminD3.this method assumes all vitaminD3 extracted. it means if you take 1 ml of plasma 1.2 ng of vitaminD3 contained.
so per 1ml of plasma we get 1.2 ng of vitaminD3 then 1.5 ml of plasma we can get 1.8ng/ml concentration.
