Mark as Read Reply The molecule of interest for my post is a family of molecules, the STAT protein family. These proteins are well known to appear in the occurrence of cancer and are the main focus of many cancer researchers. STAT3 is one of the proteins that is able to bind the cross-phosphorylated RTK's in order to drive cancer cell reproduction. The main goal of research directed at STAT3 is to inhibit its pathway in order to stop cancer cell replication. STAT proteins names are an acronym for signal transducer and activator of transcription. So, obviously you can see the importance of this protein with cancer because the expression of oncogenes (cancer-causing genes) is a root of cancer at the molecular level. When activated, the STAT3 protein is phosphorylated and a dimer forms of STAT3. This is where the first intracellular target site is for treatment as the source discusses. This is an interesting factor because as we learned in the previous week, RTK's cytoplasmic tails and proteins are bound by SH2 domain qualities and this is the exact thing that the researchers are targeting. This inhibition would not allow the STAT3 to bind the cytoplasmic tail and to make its way directly into the nucleus to affect transcription if t were successful. This is just one of many perspective sites of inhibition but was the most relatable to this course. its a very good litrature that you had mentioned about stst protein and rtk's and the study of cancer causing elements are become very useful for our whole planet because cancer is becoming very common now a days and even children are also affected form this dreaded disease.
as we know the rtk's have the critical role in development and progression of many types of cancers and mutation in it leads to activation of a series of signalling cascades which have numerous effects on protein expression.it contain a transmembrane domain.
In cancer, constitutive and aberrant activations of components of those pathways( RAS/MAP kinase and the RAS/PI3K/AKT pathways.) result in increased proliferation, survival and metastasis.
mutations affecting RTKs, Ras, B-Raf, PI3K and AKT can leads to many types of cancers that why these become prime targets for cancer therapy
