Question

1.A drug is administered to the body as a bolus injection at a dose of 100 mg. If the volume of distribution is 140 L, and the half life of the drug in the body is 3h due to urine elimination:

Determine the rate constant k

Give the drug concentration as a function of time

2.For the drug in problem (1), the therapeutic dose range is 4 to 8 g/ml. Is the 100 mg dose enough? What dose would you suggest, and how often would you give additional drug to the patient? Would you give the same dose for the subsequent injections?

Explanation / Answer

1) rate constant = ln2/half life = 0.693/3 = 0.231 hr-1

2) Injected quantity = 100 mg/140 litres = 0.714 mg/L = 0.174 g/ml.

Thus, the 100 mg dose is not enough.
