Question

Lab 12Transcription and Translation Experiment 2: Genetic Mutations For this experiment, you will be responsible for the induction of mutations in a nucleic acid sequence 1. Translate the following sequence AUG GAG GUC UUU AAG AGA CAU UUA GAU GUA GCC CUU AGU GAU GUU UAGG 2. Create one or more point mutations in this sequence AUG GAG GUC UUU AAG AGA CAU UUA GAU GUA GCC CUU AGU GAU GUU UAGG 3. Translate your mutated sequence Now create a frameshift mutation by adding one or more bases AUG GAG GUC UUU AAG AGA CAU UUA GAU GUA GCC CUU AGU GAU GUU UAGG 4. 5. Translate your mutated sequence Create a frameshift mutation by deleting one or more bases AUG GAG GUC UUU AAG AGA CAU UUA GAU GUA GCC CUU AGU GAU GUU UAGG 6. 7. Translate your mutated sequence Align the amino acid sequences that you determined for each of the four transcripts 8. Explanation / Answer

1. met-glu-val-phe-lys-arg-his-leu-asp-val-ala-leu-ser-asp-val (first codon AUG is starting codon and codes for methionone, last codon is stop codon and codes for nothing rest all amino acids are according to their codon).

2. Point mutation is a genetic mutation which causes insertion/ deletion or substitution to a single nucleotide base in a sequence of DNA or RNA.

Here i am substituting a base in second codon (GAG to GAU). here G is substituted by U which is an example of point mutation and it can be written as: AUG GAU GUC UUU AAG AGA CAU UUA GAU GUA GCC CUU AGU GAU GUU UAG.

3. met-asp-val-phe-lys-arg-his-leu-asp-val-ala-leu-ser-asp-val. (only change will occur in second amino acid because GAG codesw for glutamine while GAU codes for valine)

4. A frameshift mutation is a genetic mutation which causes insertions or deletions of a number of nucleotides in a DNA/ RNA sequence. The insertion or deletion can change the reading frame (the grouping of the codons) due to the triplet nature of codon.

Here i am inserting a base (G) just afer third codon. Now the reading frame will be: AUG GAG GUC GUU UAA GAG ACA UUU AGA UGU AGC CCU UAG UGA UGU UUA G.......

5. Translation of mutated sequence: met-glu-val-val (no further addition of amino acid will take place because UAA is stop codon). Since the whole reading frame has been changed due to single base addition.

6. Here i am deleting one base from the fourth codon (UUU to UU). Now the reading will be: AUG GAG GUC UUA AGA GAC AUU UAG AUG UAG CCC UUA GUG AUG UUU AG........

7. Translation of mutated sequence: met-glu-val-leu-arg-asp-ile. (No further addition of aminoacid will take place because UAG is stop codon which has been arises due to frameshift mutation)

8. I. met-glu-val-phe-lys-arg-his-leu-asp-val-ala-leu-ser-asp-val

II. met-asp-val-phe-lys-arg-his-leu-asp-val-ala-leu-ser-asp-val

III. met-glu-val-val

IV. met-glu-val-leu-arg-asp-ile
