Long time duration of extension stage in PCR causes the non specific amplification. And Short interval of extension time will not sufficient to replicate the gene of interest. So generally the 1min per kb time used to amplify the gene.   

As per observation there are two bands in lane-2. Lower level band is about 1.8kb (trpB).upper level band is about 2.4 kb (non specific).TrpB forward primer will bind at 3’end of the complimentary sequence. Reverse primer will bind at 5’ end of the template sequence.

In case of non specific the primer can bind at any location where it finds complimentary sequence.

This primer may be nonspecific that is amplifying the gene from different location also.
