Question

You are working on a new drug called Bioscicyn that you suspect blocks translation. You set up an in vitro translation experiment designed to produce polypeptides of 100 ammo acids After 30 minutes, 100 specifically measure the amount of protein around 100 ammo acids using a protein assay. From the data below answer the following questions. Tube 1: 100 ng mRNA, ribosomes, charged tRNAs, ATP Tube 2:100 ng mRNA, ribosome, charged tRNAs, ATP and Bioscicyn Tube 3. 100 ng mRNA, ribosome, charged tRNAs, ATP. Reaction proceeds after 15 minutes then Bioscicyn is added and reaction proceeds for 15 mins more. Why is the measurement of mRNA important In this experiment? Based on the data what process is Bioscicyn affecting? What is the mechanism of this process? Be sure to explain the results of each tube.

Explanation / Answer

Why is the measurement of mRNA important in this experiment?

DNA dose not directly get converted into protein because it converted into mRNA after that mRNA get transcribed into protein. So the amount of mRNA will define the basic source and site of protein synthesis. If the amount of mRNA is less than the conversion into protein will be low, that will not help us clearly determine the effect of bioscicyn which is playing major role in translation process inhibition. If the amount of mRNA is known then the corresponding protein synthesis can be assumed and in other hand the total loss in protein synthesis because of bioscicyn can be measured.

Based on the data what process is Bioscicyn affecting?

Bioscicyn will not cause the degradation of mRNA hence the mRNA level in second graph is stable. All three tubes have equivalent amount of mRNA. Bioscicyn majorly affects the protein synthesis. Hence the protein amount will be low in presence of bioscicyn. In case of first graph there is no second tube because it was present in start of the translation process, so it blocked the process and protein synthesis is zero. But in case of Tube-3 the bioscicyn was added after 15 min. So reaction carried for 15 minutes. SO there is small peak in comparison to tube -1 where bioscicyn is absent and high amount of protein is produced.

What is the mechanism of the process? Be sure to explain the results of each tube.

Mechanism of this experiment is based on translation process where the translation gets blocked in presence of bioscicyn. That may interact with intermediate molecules such as translation factors or path of translation. mRNA level in second image is same in all three tubes. But the amount of protein will be low in presence of Bioscicyn. Tube 1 presenting the normal process which can be used as control, But in case of tube 3 there is addition of bioscicyn after 15 min. That leads to formation of small amount of the proteins.
