Question
How much ethylenediaminetetraacetic acid (EDTA) (M.W. 372) would you use to make up 0.5 liters of a 0.5 M solution?
2. The ethidium bromide solution we will use in class is 10 mg/ml. We have added 4 µl to 100 ml of agarose gel mix.
a) How many µg of EtBr did we add to each gel?
b) What is the final concentration of EtBr in the gel?
c) How much EtBr solution at 1 mg/ml would be used to give 0.05 µg/ml EtBr final concentration? (Hint: determine dilution factor first)
d) How many molecules of EtBr are in your gels? (EtBr formula weight 394.3.)
Explanation / Answer
1)
we can solve this using Molarity formula
Molarity = (weight/ Mwt) x (1000/V in Ml)
0.5 = (weight / 372) x (1000/500ml)
weight = 93 grams required
2)
we have 10 mg for 1 ml that means
10000 micrigrams for 1000 micro liter
i just converted milligrams to micro grams and milliliter to micro liter
now 1000 micro liter have 10000 micro grams
4 micro liter will have how much
= 40000/1000
= 40 micrograms are there.
40 micro gram you have added again to 100 ml solution
40 micro gram means 4 x 10-5 grams
now concentration = grams/ml
= 4 x 10-5 / 100
= 4 x 10-5 gram/mL
remaining things i am not sure
