Question
A cysteine protease from a malaria parasite was recently isolated. A compound names P56 was found to inhibit the enzyme. Xomounds of this type have the potential to be used as drugs to treat malaria. Data were plotted as shown below.A. What kind of inhibitor is P56? B. Use the equations of the lines provides to calculate Km and Vmax for the enzyme with and without the inhibitor. C. Calculate the value of K1 in the presence of 0.22 mM inhibitor. 54. A cysteine protease from a malaria parasite was recently iso- lated. A compound named P56 was found to inhibit the enzyme. (b) Use the equations of the lines provided to calculate KM and Vma for the enzyme with and without the inhibitor. (c) Calculate the value of Ki in the presence of 0.22 mM inhibitor
Explanation / Answer
(a) The inhibitor P56 is falcipains type of inhibitor.
(b) 1/Vmax = 1/y-intercept
-1/Km = -1/x-intercept
So from the above graph we have,
Without inhibitor : y = 1.93x + 2.25
y-intercept = 1/Vmax = 2.25
Vmax = 0.444 micromol.min-1
x-intercept = 1/Km = 1.93
Km = 0.52 mg.ml-1
With inhibitor : y = 4.93x + 2.35
y-intercept = 1/Vmax = 2.35
Vmax = 0.43 micromol.min-1
x-intercept = 1/Km = 4.93
Km = 0.203 mg.ml-1
(c) With 0.22 mM of inhibitor,
K = Vmax/I = 0.444/0.22 = 2.02
