Question
Biochemistry Lab Practical:
We are to be given two pure samples to work with. A 1 mL sample of DNA and a 1 mL sample of protein. However, which sample is which is not provided. All that is given is that the protein is BSA.
What is the protcol for;
A. Figuring out which sample is the DNA and which is the protein.
B. Quantitating the concentrations of the DNA and protein samples via spectroscopy.
C. For the protein sample, we must find the concentration by two methods. First using an extinction coefficient (given) with Beer's Law. Second using a Bradford Assay to generate a standard curve.
Thanks!
Explanation / Answer
The sample with an absorption maximum at 280 nm is BSA.
The sample with an absoption maximum at 260 nm is DNA.

If the absorbance A is too high you may dilute it in an exact ratio.
2and 3. You will have given the extinction coefficients for each substance.
Use Lambert Beer
