Question
a drug is added to epidermal cell line, first cells grow in loosely coherent clusters and a week later, cell show signs of differentiating into other cell types like smooth muscle cells and neurons. If I culture this cell into commercially-available stem cell media, there is no differentiating and a clump of cells remain.
I suspect that drug may induce formation of pluropotent stem cells.
I need to know:
1-what are markers for pluropotent stem cells?
2-what experiment may be used to prove these cells are pluropotent stem cells?
3-If we feed a mice with drug, what may happen to tissue architecture if small intestine ?
Explanation / Answer
A large numner of glycoproteins and carboydrates acts as cell surface markers. For example lectins acts as markers for pluriopotent stem cells.
induced pluripotent stem cells can be generated from mouse embryonic fibroblasts and adult mouse tail-tip fibroblasts by the retrovirus-mediated transfection of four transcription factors, namely Oct3/4, Sox2, c-Myc, and Klf4 .
This drug induces the formation of pluripotent cells in the intestine. The new cells formed will be alike and they will not work as intestinal cells. These will be capable of transforming into other types of cells.
