a). Advantages to using microsatellite marker over AFLP markers are
-These markers often present high levels of inter- and intra-specific polymorphism, particularly when tandem repeats number is ten or greater.
-It is also the only molecular marker to provide clues about which alleles are more closely related. Microsatellites have proved to be versatile molecular markers, particularly for population analysis.
b). Advantages to using AFLP markers over microsatellite markers are
- AFLP not only has higher reproducibility, resolution, and sensitivity at the whole genome level compared to other techniques, but it also has the capability to amplify between 50 and 100 fragments at one time.
-No prior sequence information is needed for amplification.
5). Roche 454 Pyrosequencing is the technique used for DNA sequencing, in which the sequencing relies on the detection of pyrophosphate release on nucleotide incorporation, rather than chain termination with deoxynucleotides.
- It is also used for genome sequencing and matagenomics. It can generate 400 million nucleotide data in a 10 hour run with a single machine.
Solexa Illumina sequencing is parallel sequencing using a proprietary reversible terminator-based method that enables detection of single bases as they are incorporated into growing DNA strands.
