The repressor molecule (excessively synthesised molecule) must bind to the coreceptor molecule (buy product or end product of repressor). Then, the coreceptor activates the repressor protein, which bind to the operator site.

As trp operon works based on repressor mechanism, mutation at the operator site prevent the binding of repressor, so that the operon will never be turned off.
