the reason that you need a concentrated solution is to get agood separation. If it is dilute that molecules could juststick together and elute together. also it would be hard tosee the bands clearly to know when they were eluted. In the TLC you can compare the separation of teh compounds anduse chemical reactions or UV light to identify theunknowns. Cholesterol is more polar that cholesteryl palmitate becasueit has an OH group on the outside of the molecule the oxygens onthe cholesteryl palmitate are in the middle and have verylittle ability to interact. While ethyl acetate is more polar than dichloromethanepetroleum ether is very much less polar than dichloromethane.
